Over the past decades, it was shown that a cross-linkage of T cells with tumor cells can lead to an activation of the T cells and, as a consequence, to a destruction of the tumor cells. Such a cross-linkage can artificially be achieved by single-chain bispecific antibodies (bsAbs). In principle, they are created from the variable domains of two different monoclonal Abs: one directed against a target antigen on the surface of the tumor cell and another one directed against an activating receptor of the T cell, most commonly the CD3 complex of the T cell receptor. For redirection of T cells to hematologic malignancies and solid tumors we designed different bsAb. As underlined by in vitro and in vivo data these bsAbs are promising candidates for cancer immunotherapy. In order to further reduce the risk of side effects and accelerate the development of conventional bsAbs, we additionally developed a Lego-like modular targeting platform. For this purpose, the conventional bsAb was split into two components which are termed effector and target module. It was shown that the modular system can functionally replace conventional bsAbs both in vitro and in vivo. Due to its dual character this novel platform technology not only exhibits a higher flexibility but also opens the door for multiple as well as multifunctional tumor targeting. Overall, our bsAbs as well as the modular Ab-platform represent flexible, valuable and promising tools for cancer immunotherapy.
