- DNA is compacted inside the nucleus in the form of chromatin, in which 146 basepairs of DNA is wrapped around an octamer of histone proteins in almost two turns. In the recent years, with the advent of chromosome capture methods, it is becoming clear that chromatin folds into highly organized compartments and domains inside the nucleus. This 3D organization is often cell specific and dynamic. However, chromosome capture methods lack spatial context and typically reveal an average organization of the chromatin over millions of cells. It is therefore important to complement these methods with microscopic visualization of chromatin organization in intact nuclei to elucidate the crucial link between chromatin structure and gene activity. In my talk, I will summarize our efforts in the recent years to use quantitative super-resolution microscopy to study the organization of the chromatin in the nuclear context.
