 ation are notoriously difficult to study. We present a novel fluorescence 
 spectroscopy approach that enables continuous analysis of the interaction 
 membrane proteins with sub-microscopic clusters or the membrane cytoskelet
 on in intact cells. It quantifies the diffusion speed of proteins in live 
 cell membranes and their strength of interaction with the cytoskeleton. Re
 sults from several applications will be presented including calcium depend
 ent PIP2 modulation\, influence of general anesthetics and membrane stiffn
 ess measurements.
