- Chlamydomonas reinhardtii is the most widely used alga in basic research. Although reverse genetics via gene editing was first demonstrated in 1995, isolation of non-phenotypic mutants remained problematic due to a very low targeting efficiency. As a postdoc in the lab of Prof. Peter Hegemann (HU Berlin), Andre established Cas9 based genome editing that now enables routine genetic engineering of Chlamydomonas. Interestingly, different from many other model organisms, C. reinhardtii shows atypical gene modification preferences after Cas9 cleavage.
