In all vertebrates, early development of the oral and facial regions is generally considered well known and rather uniform. The oral-pharyngeal development comprises progressive formation of the primary mouth and pharyngeal gill openings, where the ectoderm generally outlines the outer, whereas endoderm the inner surfaces and structures. In the seminar I will discuss several advances in our understanding of the ectoderm-endodermal dynamics. The overall complexity and disparate modes of the primary mouth formation in vertebrates will be revealed and discussed. It will be shown, how the dissimilar mode of mouth formation can lead to the unexpected germ-layer origin of oral teeth. Most of the time I will discuss our recent findings on a peculiar endodermal domain - the pre-oral gut. Its contribution to oral and facial epidermis presents the first direct evidence of external surfaces and structures of the vertebrate head to be thoroughly derived from the endoderm. Aside from the primary mouth and gill slits, the pre-oral gut thus represents another ectoderm-endodermal interface, with rather unexpected cellular behaviour. The preoral gut is in most vertebrate lineages reduced, but it will be shown that it reveals an ancient blueprint of the foregut morphogenesis (and possibly also patterning) for all vertebrates.
