- Hi-C technique and microscopy together provide a comprehensive picture of spatial chromosome organization. Recent experiments and polymer modeling suggested two major mechanisms of chromosome organization. The first mechanism is the active (ATP-dependent) process of loop extrusion by SMC complexes, that is responsible for formation of domains during interphase, and chromosome compaction and segregation in metaphase. The second mechanism is the spatial compartmentalization of the genome, which we found to be mediated by affinity between heterochromatic regions. I will also discuss how loop extrusion can function as a mediator of long-range communication between genomic elements.
