In most neurodegenerative diseases certain neuronal subgroups degenerate fast while others, carrying the same mutations, subjected to theoretically analogous stress and displaying comparable functional properties remain unaffected, even at the end-stage of the disease. Why does a given neuropathology not impact all neuronal types to a similar level? Even more striking, why does it not affect all neurons of the same subtype? These are fascinating questions in neurodegeneration research that remain unanswered. Using a combination of cellular biology techniques, genome editing and single cell-omics, and utilizing brain organoids together with transgenic animal models, we investigate the molecular origin of such distinctive neuronal vulnerability. Our main goal is to switch those individual players or signaling pathways on or off to convert vulnerable neurons into resistant. Ultimately, this knowledge will allow the design of effective therapeutics targeting specific neuronal subtypes or phases of the disease.
