The PoL PhD and Postdoc seminar is a meeting where PoL early career scientists present their research to their peers. All are welcome! 

The seminar consists of two short talks by PhD students or Postdocs, each followed by a discussion in which speaker and attendants can exchange ideas, engage in scientific discussions and network with their fellow young scientists.

The talks:

  • Shivani Gunnan Ramkumar (Mateus Group) – Anisotropic shape reacquisition during pectoral fin regeneration

Upon injury, zebrafish larval pectoral fins can regenerate within 3.5 days, re-acquiring morphologies strikingly similar to uninjured controls. This project seeks to uncover the mechanisms underlying shape re-acquisition during the regeneration of pectoral fins in zebrafish larvae. We hypothesise that the shape of the fin arises from a combination of factors, including mechanical interactions within and between the tissues of the fin and directional BMP diffusion.  Our research has identified two distinct shape changes in the pectoral fin: 2D anisotropic growth- with the proximal-distal axis growing more than the anterior-posterior axis and 3D anisotropic growth marked by a transient increase in thickness. The latter is interestingly a regeneration specific phenotype. To investigate the possible contributions tothickness increase during fin regeneration, we are studying changes in individual tissue shape, cell shape, cell number and intercellular spaces using 3D volumetric tissue and cell segmentation pipelines. From this we identified that the cartilage plays a key role in the overall thickness increase of the fin. Additionally, the regenerating cartilage cells are larger and have different shapes when compared to developmental age-matched controls. This could point towards either a change in tissue mechanics or in the dynamics of BMP signaling gradients upon amputation.

 

  • Franziska Decker (Brugués Group)Bringing SCAPE 2.0 to life: Building a microscope for future biological insights

Abstract: In the Brugués lab, we use advanced fluorescence microscopy techniques to address complex biological questions. Among these, light-sheet microscopy stands out as a powerful tool for volumetric imaging of living biological samples due to its ability to minimize phototoxicity and improve imaging speed and resolution. Recently, I have constructed a SCAPE 2.0 microscope in our lab—a specialized type of light-sheet microscope that operates with an oblique plane, enabling rapid, high-resolution imaging of dynamic biological processes. In this presentation, I will talk about some technical details and unique advantages of the SCAPE 2.0 setup, including its ability to capture volumetric data with minimal sample disturbance. Additionally, I will discuss the challenges associated with the large and complex datasets it generates.
