Our group studies (i) mechanisms of cancer initiation and (ii) epigenetic mechanisms of escape from immune surveillance. To study cancer initiation we developed several murine models of retinoblastoma. We seek to understand the networks that regulate tumor susceptibility when the RB1 tumor suppressor is inactivated, and to exploit this information to prevent hereditary cancer in highly cancer-prone RB1+/- individuals. To study the epigenetics of immune escape we utilize a combination of in vitro systems and genomics, and we are particularly interested in the role of the SWI/SNF (BAF) chromatin remodeling complex and Polycomb Repressive Complex 2 (PRC2). Our goal is to understand how tumors resist immune clearance and to reactivate the dormant pathways in cancer cells.
