- The midbrain-hindbrain boundary (MHB) acts as a lineage restriction boundary and is thought to prevent the intermingling of prospective midbrain and hindbrain cells during embryonic development. The MHB also provides, as an organizer cell population, positional information to the neighbouring cells, which take different lineage decisions. The aim of the proposed project is to identify the molecular regulators that mediate cell sorting in the developing MHB. To achieve this, first, the role of differential cell-cell adhesion or surface tension in cell sorting at the MHB is studied using atomic force microscopy and live imaging techniques. Second, the molecular mechanisms controlling cell sorting and tissue border formation is addressed using zebrafish mutants of a candidate-signalling pathway. Finally, the role of cell sorting in lineage specification is addressed using lineage-tracing analysis in combination with inactivation of genes affecting cell sorting at MHB. The results obtained from this study will increase our understanding of the fundamental process of boundary formation and patterning of the embryo.
