- The dynamics of plasma membrane organization are notoriously difficult to study. We present a novel fluorescence spectroscopy approach that enables continuous analysis of the interaction membrane proteins with sub-microscopic clusters or the membrane cytoskeleton in intact cells. It quantifies the diffusion speed of proteins in live cell membranes and their strength of interaction with the cytoskeleton. Results from several applications will be presented including calcium dependent PIP2 modulation, influence of general anesthetics and membrane stiffness measurements.
