- During vertebrate development, clustered Hox genes are controlled by enhancers acting over very long distances and localized within two distinct Topologically Associating Domains (TADs) flanking the genomic cluster. In this talk I will first introduce the experimental system and then discuss two related issues concerned with this complex bimodal regulatory strategy. The first question has to do with the mechanism underlying the boundary effect, which prevents the TADs to interact either with one another, or with target genes located on either side of this boundary. The second point is concerned with the biological relevance of this boundary, i.e. its relevance during development and its function in protecting anterior structures from the dominant effect of posterior genes.
