Recent advances in regulation of Osteoclast Formation and Function

My research focused on the molecular mechanisms by which osteoclasts are generated and resorb bone. I have used extensively the power of mouse genetics to achieve these goals, taking as a starting point in most instances animals with an in vivo bone phenotype associated with osteoclast dysfunction and, wherever possible, relevant to human disease. We performed in vivo analysis of bone phenotype, complemented by in vitro culture studies of authentic osteoclast precursors, deleted of specific genes and consistently rescued the phenotype by retrovital transduction of wild type and mutant genes in null precursors. Our assays involved, micro CT, dynamic histomorphometry, dissection of signaling pathways and examination of changes in the actin cytoskeleton, all assessed in conjunction with resorption of authentic bone. These studies have led to increased understanding of how individual proteins and point mutations thereof regulation survival and proliferation of osteoclast precursors and their differentiation and function.
