- Our studies have focused upon the chromatin adjacent to the nuclear envelope (NE). Most experiments have been conducted with the human leukemia HL-60 cell line, which can be differentiated in vitro. The granulocyte forms exhibit ELCS (envelope-limited chromatin sheets) connecting the nuclear lobes. Evidence is presented that ~10 nm chromatin fibers adjacent to the NE are organized into a crisscross pattern that resembles ~30 nm fibers. Epichromatin, the surface chromatin, in all the HL-60 cell states has unique immunological properties that have been exploited for ChIP-Seq analyses. Epichromatin appears to be highly enriched in retrotransposon Alu, and exhibits spatial patterns that are chromosome specific and independent of the state of cell differentiation.
