450 Million Years of T cell Leukemia: Models and Drug Screens in Zebrafish

- The lab of Nikolaus Trede is focused on pathologic T lymphocyte development using the zebrafish model. The two main areas of study are models of T cell immunodeficiency and T acute lymphoblastic leukemia (T-ALL). The Trede lab has generated a number of mutant zebrafish lines that have defects in T cell and/or thymus development, and has mapped and positionally cloned several of these. They have also identified three mutant lines with heritable predisposition to T-ALL. These models were used to determine high-risk T-ALL signatures based on gene expression, copy number aberrations and epigenetic changes. Exploiting the versatility of our T cell reporting transgenic zebrafish line, we have also conducted a small molecule screen designed to identify new drugs with anti-T-ALL activity. This screenresulted in the identification of Lenaldekar with selective activities against hematologic malignancies.
